Rob,
Not sure what backup software you're running, but if you're running
an incremental backup, it should only pick up files which have
changed since last backup, in which case you'd be fine if all you've
done is play a file.
The other thing you could do is just leave the drive waiting to
insert another tape for a day or two (I'm not sure why you want to
span it over a few days).
